Aim: to investigate the molecular basis of oligomerisation specificity using coiled coil peptides
Different mutants of the GCN4 leucine zipper peptide have produced. I would like to investigate the effect of the mutations on (a) oligomerisation state of the peptide, (b) thermodynamic stability of the structures and (c) kinetics of the folding /unfolding reaction.
Write a research strategy to rationalise the effects of the different mutations on the leucine zipper coiled coil structure. Base the plan on at least 3 key references that reflect the experimental approach. Ideally, at least 2 papers should describe different biophysical methods. Justify the reasons for the chosen techniques; in case there can be more than one possible method of investigation.
